Sennoside, a natural dianthrone glycoside extracted from senna leaves, primarily exerts its laxative effect by being converted into the active metabolite rhein anthrone by intestinal flora in the colon.
Its mechanisms include inhibiting proximal colonic contractions to accelerate intestinal contents transport and reduce water absorption, and activating colonic macrophages to secrete prostaglandin E2, downregulating the expression of aquaporin AQP3, thereby inhibiting water transport from the intestinal lumen to the bloodstream and ultimately promoting defecation.
